Ca2+胁迫下白及种子萌发及原球茎发育过程微形态学观察

Micromorphologic Observation of Seed Germination and Procorm Development of Bletilla striata under Ca2+ Stress

  • 摘要: 以白及种子为材料,采用植物组织培养的方法,设置6个Ca2+处理浓度(0、1.65、2.47、3.30、4.12、4.95 g·L-1),研究不同浓度Ca2+胁迫对白及种子萌发及原球茎发育过程的影响。结果表明,无菌播种10 d后,2.47 g·L-1Ca2+胁迫下白及种子率先撑破种皮,无菌播种20 d后,3.3 g·L-1Ca2+胁迫下的种子叶片长势较优,无菌播种30 d后,2.47 g·L-1和3.3 g·L-1Ca2+胁迫下的种子幼叶长势较好;无菌播种15 d后,进行种子萌发率统计,Ca2+浓度为2.47 g·L-1时萌发率最高,其次是0、1.65 g·L-1浓度处理,Ca2+浓度超过2.47 g·L-1会抑制白及种子的萌发;无菌播种30 d后,4.95 g·L-1Ca2+胁迫下白及原球茎直径较大。

     

    Abstract: In this paper, the germination characteristics of Bletilla striata under six Ca2+ treatment concentrations(0,1.65,2.47,3.3,4.12,4.95 g·L-1) were studied.The effects of different Ca2+ concentrations on seed germination and procorm development were also investigated by plant tissue culture.The results showed that the seeds of B. striata first broke the seed coat under the treatment with 2.47 g·L-1 Ca2+ solution after 10 days of aseptic seeding.Additionally, seed leaves grew better under 3.3 g·L-1 Ca2+ solution after 20 d aseptic seeding, while seed young leaves grew better under 2.47 g·L-1 and 3.3 g·L-1 Ca2+ solution.After 15 d of aseptic sowing, the seed germination rate was calculated, and the highest germination rate was found when the treatment with concentration of Ca2+ was 2.47 g·L-1,followed by 0 and 1.65 g·L-1,when the concentration of Ca2+ was more than 2.47 g·L-1,the seed germination was inhibited.After 30 d of aseptic seeding, the diameter of B. striata procorm was larger under 4.95 g·L-1 Ca2+ solution.
