肉桂种子生物学特性及生活力研究
Study on Biological Characteristics and Viability of Cinnamomum cassia Seeds

摘要: 为了探究肉桂种子生物学特性及生活力测定的最适条件,对肉桂种子形态特征、千粒重、萌发特性、生活力等进行研究,探索不同萌发环境对肉桂种子发芽率的影响,并采用2,3,5-氯化三苯基氯化四氮唑(TTC)染色法测定肉桂种子生活力,通过正交设计研究不同染色温度、染色时间、TTC溶液浓度对肉桂种子生活力测定的影响。结果表明,肉桂种子平均长度为10.89 mm,平均直径为7.41 mm,千粒重为262.41 g,含水量为35.99%,在湿沙条件下发芽率最高,为76.76%;极差分析表明,TTC浓度和染色温度是影响肉桂种子生活力的较重要的因素。研究表明,湿沙催芽能显著提高肉桂种子发芽率,TTC法最佳染色条件为TTC浓度为1%、染色温度为35℃、染色时间为8 h。Abstract: In order to explore investigate the optimal conditions for the determination of biological characteristics and viability of Cinnamomum cassia seeds, morphological indexes, 1 000-seed weight, germination indexes, viability and other aspects of C. cassia seeds were studied, and the effects of different germination environment on the germination rate of C. cassia seeds were also investigated. Meanwhile, the 2,3,5-triphenyltetrazolium chloride(TTC) staining method was used to determine the viability of C. cassia seeds, and the effects of different dyeing temperature, dyeing time and TTC concentration on the viability of C. cassia seeds were further studied by orthogonal design. The results showed that the average length and average diameter of C. cassia seeds were 10.89 mm and 7.41 mm respectively, and the average of 1 000-grain weight and water content were 262.41 g and 35.99%. The highest germination rate was 76.76% under wet sand condition. In addition, range analysis showed that TTC concentration and dyeing temperature were the most important factors affecting the viability of cinnamon seeds. To sum up, wet sand could significantly improve the germination rate of C. cassia, and the best dyeing conditions of TTC method were 1% TTC concentration, dyeing for 8 h at 35 ℃.
