响应面法优化紫丁香愈伤组织诱导条件及防褐化研究

Optimization of Callus Induction Conditions and Anti-browning of Syringa oblata by Response Surface Methodology

  • 摘要: 以紫丁香的根、茎、叶和花为材料,在改良的MS+6-BA 0.5 mg·L-1+2,4-D 0.2 mg·L-1+NAA 0.2 mg·L-1培养基诱导其愈伤组织,观察愈伤组织的生长情况,筛选出最佳外植体;根据紫丁香愈伤组织诱导的情况,采用响应面的方法对6-BA、2,4-D和NAA在MS培养基中的配比进行优化;研究聚乙烯吡咯烷酮(PVP)、活性炭(AC)和柠檬酸(CA)等3种抗褐化剂对紫丁香茎愈伤组织诱导情况的影响。结果表明,紫丁香茎段是愈伤组织诱导的最佳外植体,其诱导率为83.3%;最佳培养基为MS+6-BA 0.50 mg·L-1+2,4-D 0.2 mg·L-1+NAA 0.2 mg·L-1,诱导率可达85%,且多数愈伤组织生长状况良好;0.2 g·L-1的AC对紫丁香茎愈伤组织的抗褐化效果最好,褐化率为24.5%,且愈伤组织生长状况较好。

     

    Abstract: In this study, the callus was induced from roots, stems, leaves and flowers of Syringa oblata on the modified MS medium supplemented with 0.5 mg·L-1 6-BA + 0.2 mg·L-1 2,4-D + 0.2 mg·L-1 NAA.The growth of callus was observed and the best explants were selected.The ratio of 6-BA,2,4-D and NAA in MS medium was optimized by response surface method according to the callus induction of S. oblata. Also, the effects of polyvinylpyrrolidone(PVP), activated carbon(AC) and citric acid(CA) on callus induction from S. oblata stem were studied. The results showed that the stem segment of S. oblata was the best explants for callus induction, and the induction rate was 83.3%. The optimal medium was MS + 0.50 mg·L-1 6-BA + 0.2 mg·L-1 2,4-D + 0.2 mg·L-1 NAA,the induction rate was 85%,and most of the callus grew well.The 0.2 g·L-1 AC had the best anti-browning effect on the stem callus of S. oblata,and the browning rate was 24.5%,and the callus growth was better.
