Abstract:
In this study, the callus was induced from roots, stems, leaves and flowers of Syringa oblata on the modified MS medium supplemented with 0.5 mg·L
-1 6-BA + 0.2 mg·L
-1 2,4-D + 0.2 mg·L
-1 NAA.The growth of callus was observed and the best explants were selected.The ratio of 6-BA,2,4-D and NAA in MS medium was optimized by response surface method according to the callus induction of S. oblata. Also, the effects of polyvinylpyrrolidone(PVP), activated carbon(AC) and citric acid(CA) on callus induction from S. oblata stem were studied. The results showed that the stem segment of S. oblata was the best explants for callus induction, and the induction rate was 83.3%. The optimal medium was MS + 0.50 mg·L
-1 6-BA + 0.2 mg·L
-1 2,4-D + 0.2 mg·L
-1 NAA,the induction rate was 85%,and most of the callus grew well.The 0.2 g·L
-1 AC had the best anti-browning effect on the stem callus of S. oblata,and the browning rate was 24.5%,and the callus growth was better.
