10个马铃薯新育成品种的SSR分析

SSR Analysis on Ten Newly Bred Potato Varieties

  • 摘要: 为明确内农薯1号(NNS-1)、内农薯2号(NNS-2)、内农薯3号(NNS-3)、内农薯4号(NNS-4)、内农薯5号(NNS-5)、内农薯6号(NNS-6)、红彩5号(HC-5)、紫彩1号(ZC-1)、紫彩2号(ZC-2)、紫彩3号(ZC-3)等10个马铃薯新品种在DNA分子水平上的遗传差异性,提取10个马铃薯新品种基因组DNA,并利用SSR分子标记技术对其进行了遗传差异性和聚类分析。结果表明,筛选出SSR适宜引物10对,PCR扩增实验得到多态性条带位点117个,多态性位点数占总位点的84.17%。聚类分析显示,10个马铃薯品种的遗传距离(GD值)变幅在0.126 4~0.714 3之间。以GD值0.55为基准,将10个材料划分为四类:NNS-1、NNS-2和HC-5为第一类;NNS-3、NNS-4和NNS-5为第二类;ZC-1、ZC-2和ZC-3为第三类;NNS-6为第四类;2个特异性引物S 189和S 25扩增的SSR指纹图可明确识别10个马铃薯新品种在DNA水平上的遗传差异。

     

    Abstract: To clarify the genetic differences of the ten potato varieties at DNA molecular level like Nnongshu No.1(NNS-1), Nnongshu No.2(NNS-2), Nnongshu No.3(NNS-3), Nnongshu No.4(NNS-4), Nnongshu No.5(NNS-5), Nnongshu No.6(NNS-6), Red Color No.5(HC-5), Purple color No.1(ZC-1), purple color No.2(ZC-2) and purple color 3(ZC-3) and to extract genomic DNA of ten newly bred potato varieties of which genetic difference and cluster were analysed by SSR molecular marker technique. Ten pairs of suitable SSR primers were screened out, and 117 SSR polymorphism bands were obtained by PCR amplification of genomic DNA of 10 potato varieties, with the polymorphism ratio of 84.17%.
