Abstract:
To explore the suitable culture conditions for rapid propagation of Ardisia crenata Sims embryos in vitro, the seeds of A. crenata with different maturity ages were selected as experimental materials, the regulation technology of rapid propagation and the growth of A. crenata embryos in vitro were studied. The results showed that the high germination rate of A. crenata embryos in 1/2 MS medium, was 100% after 5 days, which was significantly higher than that in MS and WPM medium(p<0.05). The germination rate of embryo increased firstly and then decreased with the increase of embryo age, proper reduction of large elements was beneficial to seedling growth, which had the best growth in 1/4 MS medium; Correlation analysis showed that transplanting survival rate was significantly positively correlated with the number of roots of seedlings(p<0.05), and was significantly negatively correlated with stem diameter of seedlings(p<0.01). Orthogonal test results showed that the optimum sucrose concentration, basic medium and plant hormone ratio could obtain the ideal plant type of with more roots and moderate root and stem diameter, and improve the survival rate of transplanting. Therefore, the regulation technology of A. crenata Sims embryo was embryo aged 47 days were incubated in 1/2 MS medium for germination and subcultured in 1/4 MS medium for growth. Strong seedlings in 1/2 MS medium with 0.5 mg/L 6-BA+2.5 mg/L NAA+20 g/L sucrose(pH=5.8), and transplanted in a mass ratio of 1∶1(nursery substrates to humus soil).
