不同方法测定花生花粉活力的比较研究

Comparative Study on Pollen Vitality Determination of Peanut by Different Methods

  • 摘要: 为筛选适合花生花粉活力快速检测的方法,采用TTC染色法、卡宝品红染色法、醋酸洋红染色法、I2-KI染色法、红墨水染色法以及离体萌发法等6种方法测定花育23、四粒红和吉花23的花粉活力。结果表明,不同测定方法间存在显著差异(p<0.05),TTC染色法不能使花粉着色,测定结果均为0;卡宝品红染色法和醋酸洋红染色法测定结果显著高于其他测定方法(p<0.05);离体萌发法直观准确,但操作复杂、费时;I2-KI染色法和红墨水染色法操作简单、染色效果明显,是较好的花生花粉活力快速检测方法,其中红墨水染色法的测定结果最接近离体萌发法。综合比较6种测定方法,红墨水染色法可用于花生花粉活力的快速检测。

     

    Abstract: To screen suitable methods for rapid detection of peanut pollen vitality, the pollen vitality of Huayu 23, Silihong, and Jihua 23 were determined by TTC staining, carbol-fuchsin staining, acetocarmine staining, I2-KI staining, red ink staining, and in vitro germination method. The results showed that there were significant difference(p<0.05) between the different determination methods. TTC staining could not color the pollen and the measured values were all 0;The determination results of carbol-fuchsin staining and acetocarmine staining were significantly higher than that of other determination methods(p<0.05); The in vitro germination method was intuitive and accurate, but the operation was complicated and time-consuming; I2-KI staining and red ink staining were simple and effective methods for rapid detection of peanut pollen vitality, of which the results of red ink staining were closest to that of in vitro germination. Comprehensively compared with six determination methods, the red ink staining method could be used for the rapid detection of peanut pollen vitality.
