To screen suitable methods for rapid detection of peanut pollen vitality, the pollen vitality of Huayu 23, Silihong, and Jihua 23 were determined by TTC staining, carbol-fuchsin staining, acetocarmine staining, I
2-KI staining, red ink staining, and in vitro germination method. The results showed that there were significant difference(p<0.05) between the different determination methods. TTC staining could not color the pollen and the measured values were all 0;The determination results of carbol-fuchsin staining and acetocarmine staining were significantly higher than that of other determination methods(p<0.05); The in vitro germination method was intuitive and accurate, but the operation was complicated and time-consuming; I
2-KI staining and red ink staining were simple and effective methods for rapid detection of peanut pollen vitality, of which the results of red ink staining were closest to that of in vitro germination. Comprehensively compared with six determination methods, the red ink staining method could be used for the rapid detection of peanut pollen vitality.
