多肉植物“麦秆22号”离体培养体系建立

Establishment of Culture in Vitro System of Succulent Plant Maigan 22

  • 摘要: 以多肉植物“麦秆22号”为试验材料,研究外植体类型、激素配比对“麦秆22号”愈伤组织诱导及离体培养的影响,以实现对“麦秆22号”离体培养条件的优化,为建立工厂化育苗技术体系提供参考。结果表明,以“麦秆22号”的叶片切片作为外植体诱导愈伤组织效果最佳,茎切片作为外植体诱导不定芽效果最佳。愈伤组织最佳增殖培养基为MS+2.0 mg/L6-BA+0.5 mg/L NAA+7%琼脂+30%蔗糖;不定芽最佳增殖培养基为MS+0.5 mg/L 6-BA+0.5 mg/L NAA+7%琼脂+30%蔗糖;生根最佳培养基为MS+0.2 mg/L 6-BA+0.1 mg/L NAA+7%琼脂+30%蔗糖。

     

    Abstract: To optimize the tissue culture conditions of Maigan 22 and establish a factory seedling technology system, succulent plant Maigan 22 was as material to study the effects of explants type and hormone ratio on callus induction and culture in vitro. The results showed that the best explants for callus induction and adventitious bud induction was the leaf section and stem section of Maigan 22, respectively. The optimal medium for callus proliferation was MS+2.0 mg/L 6-BA+0.5 mg/L NAA+7% agar + 30% sucrose. The optimal medium for adventitious bud proliferation was MS+0.5 mg/L 6-BA+0.5 mg/L NAA+7% agar + 30% sucrose. The optimal rooting medium was MS+0.2 mg/L 6-BA+0.1 mg/L NAA+7% agar + 30% sucrose.
