26个香菇菌株的遗传多样性ISSR分析

ISSR Analysis of Genetic Diversity of 26 Lentinula edodes Strains

  • 摘要: 为明确保存香菇种质资源之间的亲缘关系,为选育具有自主知识产权、适应内蒙古西部地区栽培的新品种提供依据。采用ISSR分子标记,用10条引物对26个香菇菌株进行多态性检测,用NTSYS软件进行聚类分析和遗传多样性比较。结果表明,10对引物能够扩增出70条稳定的条带,多态性条带67条,多态性比率为95.71%,遗传距离变幅介于0~0.749 9之间。在遗传相似性系数为0.678时,菌株划分为4个类群。ISSR分子标记技术可有效揭示26个香菇菌株种质资源的遗传多样性和品种间的亲缘关系。

     

    Abstract: In order to clearly preserve the relationship between the germplasm resources of Lentinula edodes, the new varieties with independent intellectual property rights and suitable for cultivation in western Inner Mongolia were selected to provide a basis. The polymorphism of 26 Lentinula edodes isolates was detected by using ISSR molecular marker and 10 primers. Cluster analysis and genetic diversity comparison were performed by NTSYS software. The results showed that 70 stable bands and 67 polymorphic bands could be amplified with 10 pairs of primers, the polymorphism ratio was 95.71%, and the range of genetic distance ranged from 0 to 0.749 9. When the genetic similarity coefficient was 0.678, the strains were divided into 4 groups. ISSR molecular marker technique could effectively reveal the genetic diversity of germplasm resources of 26 Lentinula edodes strains and the relationship between them.
