2个佛手瓜品种核型分析及基因组大小测定

Karyotyping Analysis and Genome Size Determination of Two Sechium edule Varieties

  • 摘要: 本试验以贵州省惠水县主要品种绿皮佛手瓜、白皮佛手瓜为材料,通过去壁低渗法染色体制片、流式细胞术分别对其核型、基因组大小,染色体数量、倍性等特征进行研究。结果表明,贵州惠水白皮佛手瓜、绿皮佛手瓜均为2倍体,染色体数目均为28条,白皮佛手瓜、绿皮佛手瓜的核型公式均为2n=2x=28m,白皮佛手瓜核型类型为1B型,绿皮佛手瓜为1C型,绿皮佛手瓜的不对称系数为52.38%,白皮佛手瓜的不对称系数为52.07%,通过流式细胞术测定,白皮佛手瓜、绿皮佛手瓜基因组大小分别为(0.73±0.02)Gb、(0.70±0.02)Gb。

     

    Abstract: In this study, the karyotype, genome size, chromosome number and ploidy of the main varieties of green and white Sechium edule in Huishui County, Guizhou Province were studied by chromosome dissection and flow cytometry. The results showed that both white-skinned and green-skinned Sechium edule were diploid and had 28 chromosomes. The karyotype formulas of white-skinned Sechium edule and green-skinned Sechium edule were 2n=2x=28m, the karyotype of white-skinned Sechium edule was type 1B, and that of green-skinned Sechium edule was type 1C. The asymmetry coefficient of green-skinned Sechium edule was 52.38%, and the asymmetry coefficient of white-skinned Sechium edule was 52.07%, by flow cytometry, the genome size of white-skinned and green-skinned Sechium edule was(0.73±0.02) Gb and(0.70±0.02) Gb, respectively.
