Genetic Diversity Analysis of Native Walnut Germplasms in Guizhou by SRAP Marker

  • Genetic diversity of the 168 native walnut germplasms collected from Panxian, Pu’an, Guiyang, Xingyi, Bijie, Weining and Yanhe in Guizhou was analyzed using SRAP marker. The results showed that 19 pairs of primer screened out from 132 pairs of primer combinations could amplify clear, rich, and good repetitive bands, and a total of 309 clear and repeatable bands were amplified from 168 samples using these primer combinations. Among them, 281 bands were polymorphic, and the polymorphism ratio was 90.94%. The results of cluster analysis showed that at the similarity coefficient of 0.62, all walnut germplasms were clustered into two groups, while at the similarity coefficient of 0.72, these walnut germplasms were clustered into ten groups, which indicated that these native walnut germplasms were related to each other. Additionally, the walnut germplasm from Xingyi had the highest level of genetic diversity among the germplasms studied.
