Study on Tissue Culture and Rapid Propagation of Spathiphyllum patinii

  • In this study, the technique of tissue culture and rapid propagation of Spathiphyllum patinii was studied by using young stem segment as experimental material. The results showed that the best disinfection methods were as follows water washing for 2 h, 0.05% NaClO treatment for 20 min, 75% alcohol treatment for 20 min and 0.1% HgCl2 treatment for 15 min after being transferred to the ultra-clean worktable. The optimum conditions for adventitious bud induction were MS medium+3 mg·L-1 6-BA+0.05 mg·L-1 NAA, illumination 2 000 lx, temperature 25—28 ℃. The maximum adventitious bud proliferation coefficient was obtained under the conditions of MS+0.2 mg·L-1 KT+2 mg·L-1 6-BA+0.2 mg·L-1 NAA at 28 ℃. The maximum adventitious shoot height was obtained on MS medium+1 mg·L-1 6-BA+0.3 mg·L-1 NAA at 28 ℃. The rooting rate of 2—4 cm seedlings was 100% when transplanted into 1/2 MS medium with 0.2 mg·L-1 NAA and 0.2 mg·L-1 IBA.
