Determination and ISSR Analysis on Polysaccharide Content in Different Germplasm Resources of Polygonatum odoratum

Abstract
To clarify the difference of polysaccharide content and genetic similarity of different Polygonatum odoratum germplasm resources, the polysaccharide content of 17 Polygonatum odoratum germplasm resources was detected by UV-vis spectrophotometry, and the genetic structure of Polygonatum odoratum was analyzed by ISSR molecular marker method. The results showed that the polysaccharide content of 17 Polygonatum odoratum germplasm resources was from 6.12% to 9.33%. There were significant differences in the polysaccharide content of 17 Polygonatum odoratum germplasm resources. At the 1% significant level, there were significant differences in all except No.1 and No.5. The genetic similarity of 17 germplasm resources of Polygonatum odoratum ranged from 0.163 8 to 0.923 0. By constructing phylogenetic tree, 17 Polygonatum odoratum were divided into three categories at the genetic similarity coefficient of 0.56. The study made it clear that there was a certain correlation between the genetic relationship and geographical distribution of Polygonatum odoratum germplasm resources, and there was no obvious relationship between Polygonatum odoratum polysaccharide content and geographical source.
