Establishment of Real-time Quantitative PCR Detection Method for Five Potato Viruses

Abstract
To improve the detection efficiency and throughput of potato virus, enzyme-linked immunosorbent assay(ELISA) and real-time PCR(qRT-PCR) were used in this study to detect X virus, Y virus, S virus, leaf curl virus and spindle tuber virus in potato seedlings, in order to construct the detection method for five kinds of potato viruses. The results showed that the positive detection rate of qRT-PCR was 6%, and the sensitivity was higher than that of DAS-ELISA. The qRT-PCR method had strong specificity, good repeatability and high sensitivity, and could effectively detect five kinds of potato viruses.
