Abstract:
In order to assess the effects of cutting position, substrate, growth regulator on rooting rate and quality of cuttings of Thuja sutchuenensis Franch., and to find the optimal treatment scheme to improve the rooting rate and root quality of cuttings, the orthogonal table L
9(3~4) was used to carry out the cutting experiment with the factors of substrates(peat-vermiculite-perlite, edible fungus residue-vermiculite-perlite, and carbonized rice husk-vermiculite-perlite, three mixed substrates were prepared according to the volume ratio of 1∶1∶1), positions(upper, middle and lower part of the canopy of T. sutchuenensis), and treatments of growth regulator(growth regulator ABT-1 with concentrations of 125, 500 and 1 000 mg/L, and soaking time of 8 h, 2 h and 2 min, respectively). The rooting rate and root quality of cuttings were investigated after 210 days. The results showed that the rooting rates of cuttings were 90%, 88% and 41% on peat-perlite-vermiculite, edible fungus residue-perlite-vermiculite and carbonized rice husk-perlite-vermiculite, respectively, with variance analysis showed a significant difference(P <0.05). Analysis of variance also showed that the number of primary and secondary adventitious roots, the longest root length and diameter, and root biomass of cuttings were significantly different(P <0.05) on different substrates. There was no significant difference in rooting rate and root development indexes of cuttings from different positions and growth regulator treatments. In general, position of cuttings, substrate, growth regulator treatment all had certain effects on the rooting rate and development of cuttings of T. sutchuenensis, but the influence of the substrate is greater, and the cutting position and growth regulators treatment have less. Therefore, the best treatment to improve the rooting rate and root quality of cuttings of T. sutchuenensis was to collect them from the lower part of the canopy of the mother tree, soak the cuttings in ABT-1 solution at a concentration of 1 000 mg/L for 2 min, and use the mixed substrate of peat(edible fungus residue)-perlite-vermiculite.
